ITO (Indium Tin Oxide) and FTO coated glass slides provided from Nanocs were made by coating ITO or FTO to one side of soda lime glass. These ITO and/or FTO coated glass, also called transparent conductive glass (TCO), come with different surface resistivity from 5 ohm/sq to 100 ohm/sq, with thickness from 0.5 mm to 2.2 mm. ITO and FTO coated glass has been used in LCD/OLED devices and many other devices requiring high electrical conductivity and good optical transmission. 酪胺信号放大技术(Tyramide Signal Amplification)又称催化信号放大技术(Catalyzed Signal Amplification),是一类利用HRP对靶抗原进行高密度原位标记的酶学检测方法,其不但可以用于IF/IHC的信号放大,亦可用于Elisa、ISH等检测。
酪胺信号放大技术可以用于检测用传统方法无法检出的低丰度靶标。基于酪胺的信号放大技术能够提供极强的灵敏度、检测极微量的目的抗原。酪胺信号放大技术极大的降低抗体的用量,节约抗体。酪胺信号放大试剂盒可与传统染色方法结合使用以多色成像,也可以顺序进行两个或更多个酪胺反应以标记一个样品上的不同靶标。

注意事项
1. 使用前请将产品瞬时离心至管底,再进行后续实验。
2. 1×Tyramide Amplification Buffer首次使用后,建议小量分装,-20℃保存,避免反复冻融。
3. 与荧光二抗相比,Tyramide试剂盒显示出更高的灵敏度和更强的信号,因此,实验时一抗的使用浓度较低,建议梯度设置一抗浓度以找到最佳浓度。
4. 建议设置未孵育一抗的阴性对照,确保阴性对照在孵育和洗涤过程中没有被阳性样品中的试剂交叉污染。对于组织样品,建议对未染色的对照(不添加抗体或酪酰胺)进行成像,确定组织是否有自发荧光,排除对背景的影响。
5. 建议使用5 μg/mLHRP结合物,降低浓度可能会影响信号强度和灵敏度。
6. 建议1:200稀释YF®/Biotin-Tyramide。较高的浓度可能会导致信号过强或背景高,建议从1:1001:1000梯度稀释。
7. 可依次使用多个YF®/Biotin-Tyramide来标记同一样品的不同靶标,每次酪酰胺反应后需进行HRP淬灭或抗体剥离。
8. 多色标记时,依据抗原密度不同选择不同荧光素(低密度抗原选择强染料;高密度抗原选择较弱染料)。标记顺序对最终标记效果有一定影响,需自行摸索。

什么是TSA技术?

TSA技术,即酪酰胺信号放大技术(Tyramide signal amplification, TSA)是一类利用辣根过氧化酶(HRP)对靶蛋白或核酸进行高密度原位标记的酶学检测方法。

TSA试剂盒有哪些组分?
试剂盒固定组分BSA、1×Tyramide Amplification Buffer,可变组分Tyramide Stock Solution, 200×、HRP-conjugated secondary antibody。Tyramide Stock Solution, 200×可根据需求选择不同波长的YF/Biotin染料。HRP-conjugated secondary antibody可根据一抗在Goat anti-mouse IgG、Goat anti-rabbit IgG、Streptavidin中进行三选一。

TSA试剂盒适用于哪些样本?
TSA试剂盒适用于细胞、石蜡切片、冰冻切片样本。

石蜡切片样本进行多色标记的步骤?
每一轮标记后执行微波处理(抗原修复步骤)以去除抗体,再接下一轮的标记,因此可选择同种属来源的一抗,二抗具有通用性。

细胞样本可以用TSA技术进行多色标记吗?
不推荐。若需要,建议使用细胞爬片或细胞涂片进行,并且抗体剥离及抗原修复推荐使用商业化的抗体剥离液,尽管如此,经过几轮染色清洗后,细胞流失仍会较多,可能不利于后续结果的分析,具体可根据需求尝试。
