产品介绍

DiO Plus效能等同于Neuro-DiO。
DiO Plus是一种代替膜绿色荧光染料DiO的新型染料，DiO由于其溶解度低、易形成聚合物和横向扩散速度缓慢等缺点，很难用于神经元和细胞悬液的研究。DiO Plus和DiO具有几乎相同的吸收光谱和发射光谱，但前者具有更好的膜溶解性，并且不形成非荧光性的聚集体，通常这也会减慢染料在膜上的扩散速率。
DiO Plus染色后可进行多聚甲醛（不可使用甲醇等其他试剂）的固定，但不建议在染色后进行透化的过程。此外，在固定透化（室温下用0.1% TritonX-100 透化）后，也可以很好地进行质膜染色。
以每次使用100 μL染色工作液，染色工作液浓度3 μM计算，10 mg配置为工作液大概可以用15346次。

注意事项
1. 使用前请将产品瞬时离心至管底，再进行后续实验。
2. DiO Plus染色固定的细胞或组织样品时，通常使用配制在PBS中的4%多聚甲醛进行固定，使用其它不适当的固定液会导致荧光背景较高。
3. 荧光染料均存在淬灭问题，请尽量注意避光，以减缓荧光淬灭。

◆相较于DiO染料，DiO plus的优势是什么？

DiO plus具有更好的膜溶解性，并且不形成非荧光性的聚集体，在膜上的扩散速度更快。

◆细胞追踪有很多产品，该如何选择？
首先确定追踪细胞的时间，然后考虑染料结合机制。
1.钙黄绿素染料（C4041）标记均匀且对短期细胞迁移示踪效果极佳，但也会被某些类型细胞迅速外排。
2.亲脂性的花青素染料，如DiI（D4010），DIO（D4007），DiA(D4059)，DiD(D4019)，DiR(D4006)能标记细胞膜而不破坏其功能，并且能持续更长时间，但如果发生膜融合则可能会染上其他细胞。此外，它们还会在透化过程中丢失。
3.CellTracker染料（C4060）更有利于长期标记，其带有温和的氯甲基反应基团使之能够与细胞组分共价结合。
4.CFDA SE（C4070）也能共价地结合于细胞组分。
在所有列出的试剂中，细胞内保留与否取决于细胞分裂的速率和细胞的固有特性（主动外排，膜和蛋白质的周转率等）。其中共价结合试剂比非共价结合的试剂展现出更长的保留时间。

烟酰胺腺嘌呤二核苷酸(Nicotinamide adenine dinucleotide, NAD)是所有细胞中都存在的一种辅酶，包括NAD+(氧化型)和NADH(还原型)两种形式。NAD+既是氧化还原反应过程中传递电子的辅酶，又可以作为很多酶的底物来参与细胞内反应。NAD+在细胞和体内发挥着重要的功能，其合成和降解及其产物参与细胞凋亡、代谢调控和基因表达的调控等，并且NAD+的减少是细胞死亡的主要因素之一。NAD+在调节细胞氧化还原状态方面的重要性以及调控信号通路及转录方面的功能，使得NAD+及其合成和消耗的酶成为多种疾病的潜在药物靶点。传统的NAD+/NADH和NADP+/NADPH测定是通过检测340 nm处的吸收来完成的，该方法灵敏度低且易受干扰。NAD/NADH检测试剂盒是一种基于WST-8的显色反应，通过比色法来检测细胞、组织或其它样品中NAD+(氧化型辅酶Ⅰ)和NADH(还原型辅酶Ⅰ)各自的量、比值和总量的检测试剂盒。NAD/NADH检测试剂盒能特异性地检测NAD+和NADH，而不检测NADP+和NADPH，在反应过程中NAD+被还原为NADH，NADH将WST-8还原成橙黄色formazan（甲臜），在450 nm左右有最大吸收峰。反应体系中生成的formazan与样品中NAD+或NADH的总量呈比例关系。本试剂盒可检测0.1 μM-10 μM NAD+或NADH。

注意事项
1. 建议NADH标准品溶解后小量分装保存，避免反复冻融。
2. NAD+/NADH提取液粘稠，使用过程中务必保证和待加入的体系充分混匀。
3. 加样和混匀过程中，应避免产生气泡影响最终的吸光度检测。
4. 由于NAD+/NADH不稳定，易降解，所以尽量使用新鲜样品进行检测。

特性

 在免疫沉淀实验流程中富集经 m6A修饰的 RNA
 经富集的 RNA 可直接用于 NGS 或RT-qPCR

概述

 EpiMark® N6-甲基腺嘌呤富集试剂盒包含一个 N6-甲基腺苷（m6A）兔源单克隆抗体。试剂盒中含有2个对照 RNA，一个含有 m6A 修饰（Gaussia 荧光素酶），另一个不含 m6A 修饰 (Cypridina 荧光素酶)用于监测富集和消耗程度。其中，Gluc RNA 对照在转录过程中需 20% 的 m6ATP 和 80% 的 ATP。
该试剂盒可用在免疫沉淀实验流程中富集经 m6A 修饰的 RNA，可用于 NGS 或 RT-qPCR 等下游实验。在片段化的 RNA 样本中，经修饰的 RNA 可与 N6-甲基腺苷抗体结合，从而被于抗体结合的 Protein G 磁珠富集。再经多轮洗涤与纯化步骤后，富集的 RNA 洗脱于无核酸酶水中，可用于进一步的分析。

Description:

Azide/azido functionalized polyethylene glycol (N3-PEG-X) is a bifunctional PEG derivative that can be used to modify proteins, peptides and other materials. Azide group can react with alkyne in aqueous solution catalyzed by copper. It can also be reduced into amine group easily while another functional group can be react with other molecules or functional groups. PEGylation can increase solubility and stability and reduce immunogenicity of peptides and proteins. It can also suppress the non-specific binding of charged molecules to the modified surfaces.

Physical Properties:

  • Off-white/white solid or viscous liquid depends on molecule weight;
  • Soluble in regular aqeous solution as well as most organic solvents;
