        凝集素(Lectin)是指一种从各种植物,无脊椎动物和高等动物中提纯的糖蛋白或结合糖的蛋白,因其能凝集红血球(含血型物质),故名凝集素。常用的为植物凝集素(Phytoagglutin,PNA),通常以其被提取的植物命名,如刀豆素A(Conconvalina,ConA)、麦胚素(Wheat germ agglutinin,WGA)、花生凝集素(Pehaiut agglutinin,PNA)和大豆凝集素(Soybehai agglutinin,SBA)等,凝集素是它们的总称。

通过盐析法、离子交换层析、凝胶过滤、亲和层析一系列操作对提取物进行分级分离,筛选富集对H5N1具有最强微中和活性的组分,通过电泳发现为包含五种肽段的寡聚蛋白,质谱分析显示该蛋白质为扁豆凝集素(FRIL)。已有研究表明扁豆凝集素单体具有一个糖结合结构域,但在溶液中的状态未得到确定,综合利用多角度光散射、动态光散射、电镜复染,发现扁豆凝集素为四聚体结构。

进一步在体外定量测定了扁豆凝集素的抗病毒活性,空板减少试验与微中和试验显示PRNT50EC500.7ug/Ml。利用11种流感毒株测定中和谱,发现除实验室菌株A,扁豆凝集素能以纳摩尔水平抑制不同亚型毒株。研究者通过鼻腔给药在小鼠体内评估凝集素的作用,凝集素抗病毒活性表现剂量依赖,高剂量给药能够明显延长存活时间甚至保护免受感染。

扁豆凝集素抗病毒活性

扁豆凝集素是已知的甘露糖/葡萄糖特异性凝集素,但关于扁豆凝集素与复杂糖基化的结合机制还有待了解。研究者将接种流感病毒的鸡胚进行α甘露糖酶抑制剂并在回收时利用切割高甘露糖的内切糖苷酶处理,纯化后利用串联质谱、免疫印迹分析。

 结果发现扁豆凝集素主要结合流感病毒血凝素,而且主要结合复合型糖链而不是高甘露糖链,这与其他凝集素有很大不同。为了探明这现象背后的机制,研究者利用荧光探针标记的扁豆凝集素,发现与1,31,4岩藻糖基化N乙酰葡萄胺的复合型糖链结合好。

凝集素结合糖链

进一步分析了扁豆凝集素的抗病毒机制,竞争性微中和试验发现甲基甘露糖苷和葡萄糖能够抑制其中和作用,证实了抗病毒中和效应依赖于糖结合功能。细胞用扁豆凝集素、流感病毒依次分开处理,不能有效中和病毒感染,说明扁豆凝集素抑制病毒进入,必须直接结合病毒粒子才能发挥作用。流感病毒侵入细胞主要是吸附、内吞、脱壳、核转运这几个步骤,通过血凝抑制试验发现扁豆凝集素不能抑制病毒的吸附,研究者通过低温减慢病毒内吞入胞,使病毒吸附在细胞上,加入扁豆凝集素后观察到明显的中和效果,而在常温下病毒发生内吞后无法发挥作用,因此凝集素作用于吸附与内吞之间的过程。利用病毒核蛋白抗体免疫荧光示踪病毒入胞过程,发现凝集素处理组导致病毒滞留在核周区,通过内体/溶酶体染色显示病毒停留在晚期溶酶体内,凝集素在溶酶体仍能保持活性,可能发挥阻止病毒核转运的作用。进一步研究者发现核蛋白免疫荧光表现为大颗粒,且随着凝集素浓度增加颗粒逐渐变大,推测是凝集素与病毒颗粒间形成多价交联,聚集成团,从而发挥抗病毒作用。
