Super Safe Green 核酸染料, 10,000× in DMSO是一款新型核酸染料(效能等同于赛默飞家的SYBR Safe),其主要成分是花菁染料。该染料具有高灵敏、高信噪比、高稳定性、无致突变性超安全(在工作浓度中)、适用范围广、仪器兼容性好的特点。可用于各种核酸电泳及各种片段大小的染色,同时兼容标准凝胶成像系统和可见光激发的凝胶观察装置,适用于紫外凝胶成像系统或蓝色可见光激发的凝胶观察装置。

注意事项

1. 使用前请将产品瞬时离心至管底,再进行后续实验。

2. 如果条带总是弥散或分离不理想,请使用泡染法染色以确认问题是否与染料有关。如果染色后问题依旧存在,则说明问题与染料无关,请尝试:降低琼脂糖浓度;选用更长的凝胶;延长凝胶时间以保证边缘清晰;改进上样技巧或选择泡染法染色。

3. 该染料对玻璃器皿和非聚丙烯材料具有一定的亲合力。建议在稀释、贮存、染色等使用过程中用聚丙烯类容器。对于聚丙烯酰胺凝胶请使用泡染法。

概述

NEB 提供大量的用于标记 SNAP-、CLIP- 和 ACP-tag 融合蛋白的荧光底物供研究者选择。SNAP-tag 底物是由染料及通过苄基键耦联到染料上的鸟嘌呤或氯嘧啶构成;CLIP-tag 底物是由染料及通过苄基耦联至染料上的胞嘧啶构成。这些底物无需酶促反应,即可自我标记相应的 tag 标签。细胞通透性底物(SNAP- 和 CLIP-Cell)不仅能标记细胞内部也能标记细胞表面,而细胞非通透性底物(SNAP- 和 CLIP-Surface)仅能特异性的标记在细胞表面表达的融合蛋白。
 
ACP-tag 的标记基团与辅酶 A(CoA)的磷酸泛酰巯基乙胺耦联,在 ACP 或 SFP 合成酶作用下,完成标记反应。标记反应仅特异地发生在细胞表面表达的融合蛋白上。使用同种底物标记 ACP-tags;用于标记的合成酶标记决定反应特异性(见 292 页)。
