TCO PEG Fluorescein, also termed as TCO PEG FITC is a fluorescentPEG TCO derivative that gives strong green fluorescence. TCO (transcyclooctene) group reacts with tetrazine group fast and efficiently while fluorescein is a strong green fluorescent molecule that has excitation/emission wavelength around 495/515 nm. Fluorescein can be easily detected by fluorescent instruments at FITC channel. The reaction of TCO and tetrazine is a bioorthogaonal reaction that enables the conjugation of two molecules in aqueous solution fast and efficiently. PEG TCO derivatives have excellent water solubility, possess fast kinetics and stability in aqueous buffer. 概述

NEB 提供大量的用于标记 SNAP-、CLIP- 和 ACP-tag 融合蛋白的荧光底物供研究者选择。SNAP-tag 底物是由染料及通过苄基键耦联到染料上的鸟嘌呤或氯嘧啶构成;CLIP-tag 底物是由染料及通过苄基耦联至染料上的胞嘧啶构成。这些底物无需酶促反应,即可自我标记相应的 tag 标签。细胞通透性底物(SNAP- 和 CLIP-Cell)不仅能标记细胞内部也能标记细胞表面,而细胞非通透性底物(SNAP- 和 CLIP-Surface)仅能特异性的标记在细胞表面表达的融合蛋白。
 
ACP-tag 的标记基团与辅酶 A(CoA)的磷酸泛酰巯基乙胺耦联,在 ACP 或 SFP 合成酶作用下,完成标记反应。标记反应仅特异地发生在细胞表面表达的融合蛋白上。使用同种底物标记 ACP-tags;用于标记的合成酶标记决定反应特异性(见 292 页)。
