ADC(抗体药物偶联物)的组成条件及基本原理
ADC由以下三部分组成:
1 抗体(haitibody):其作用是特异性识别靶细胞
2 毒素(drug):起到*伤细胞的作用
2 连接物(linker):连接抗体与毒素
其实简单来看,ADC就是单克隆抗体与化疗的一种组合疗法,既具有单抗的选择性,又具有化疗的*伤力。在患者接受ADC治疗之后,ADC将会进入患者的体内循环,一旦ADC的抗体部分特异性地结合到肿瘤细胞表面,肿瘤细胞的细胞膜将会向内凹陷,形成内体囊泡(endosome)将ADC吞入,这个过程又称为内化(internaliztion);ADC的抗体部分或linker在细胞内往往会被一些酶类降解,从而将毒素释放出来,释放出的毒素便开始发挥作用,*伤细胞(不同的毒素*伤细胞原理不同,但大多数毒素是在细胞核内发挥功能)。如下图所示:
