抗体偶联药物ADC”如何消灭肿瘤细胞

ADCs是单克隆抗体和细胞毒分子的偶联物,兼具抗体的高选择性、较好的稳定性及药代动力学特征和细胞毒分子的高毒性。

ADCs可分为三部分: 单克隆抗体( monocl onal haiti bodi esmAbs) 、细胞毒分子( warhead or payl oad) 和连接基( 1i nker)

ADCs对肿瘤细胞的作用机制

ADCs以静脉注射的方式进入血液循环, 能避免被胃酸和蛋白水解酶破坏。 通过血液循环进入肿瘤组织后, 与肿瘤细胞表面的抗原结合,在受体介导下通过网格蛋白( clathrin) 的牵引发生细胞内吞, 形成核内体,氢离子流入核内体中形成酸性环境, 有利于新生的Fc受体与ADCsmAbs部分结合, 其中部分ADCsFcRns结合后被排出细胞外, 在碱性环境下, 释放出ADCs, 重新进入体循环,随后核内体与溶酶体发生融合, 未被排出细胞外的ADCs在溶酶体的降解下,释放细胞毒分子到细胞质中。 细胞毒分子与DNA、微管蛋白等结合后, 影响其复制或有丝分裂, 导致细胞凋亡。

ADCs的出现解决了传统细胞*药物难以进行靶向性治疗的问题, 具有应用前景。 随着一系列ADCs进入临床和上市, 为该类药物研发提供了方向: 增强药物靶向能力、 提高药物在血浆中的稳定性、 降低耐药性等。
