抗体偶联药物“ADC”如何消灭肿瘤细胞
ADCs是单克隆抗体和细胞毒分子的偶联物,兼具抗体的高选择性、较好的稳定性及药代动力学特征和细胞毒分子的高毒性。
ADCs可分为三部分: 单克隆抗体( monocl onal haiti bodi es,mAbs) 、细胞毒分子( warhead or payl oad) 和连接基( 1i nker) 。
ADCs对肿瘤细胞的作用机制
ADCs以静脉注射的方式进入血液循环, 能避免被胃酸和蛋白水解酶破坏。 通过血液循环进入肿瘤组织后, 与肿瘤细胞表面的抗原结合,在受体介导下通过网格蛋白( clathrin) 的牵引发生细胞内吞, 形成核内体,氢离子流入核内体中形成酸性环境, 有利于新生的Fc受体与ADCs的mAbs部分结合, 其中部分ADCs与FcRns结合后被排出细胞外, 在碱性环境下, 释放出ADCs, 重新进入体循环,随后核内体与溶酶体发生融合, 未被排出细胞外的ADCs在溶酶体的降解下,释放细胞毒分子到细胞质中。 细胞毒分子与DNA、微管蛋白等结合后, 影响其复制或有丝分裂, 导致细胞凋亡。
目前,已上市四种“ADC”抗体药物偶连物如下图所示:
ADCs的出现解决了传统细胞*药物难以进行靶向性治疗的问题, 具有应用前景。 随着一系列ADCs进入临床和上市, 为该类药物研发提供了方向: 增强药物靶向能力、 提高药物在血浆中的稳定性、 降低耐药性等。
